菠菜叶绿素的提取优化及其酪蛋白分散体系的研究

中文摘要:叶绿素是一种天然的色素添加剂,但因其疏水性及加工与贮存条件下的不稳定性在食品工业中的应用受到了限制。因此,本研究在优化菠菜叶绿素提取工艺的基础上,利用酪蛋白酸钠的双亲特性,采用反式溶剂法将菠菜叶绿素包封于酪蛋白分散体系中,并评价叶绿素和叶绿素-酪蛋白分散体系的稳定性。结果表明,以无水乙醇为提取溶剂,提取温度50 ℃,超声波300 W,提取时间30 min,叶绿素提取量可达最大值1.89 mg/g;酪蛋白可减少叶绿素的光降解,提高叶绿素溶液颜色稳定性,含有等浓度叶绿素的乙醇溶液和酪蛋白溶液经过光照(9 000 lx)处理后的叶绿素保留率分别是56.07%和62.96%;同等处理条件下,叶绿素-酪蛋白酸钠溶液总色差(ΔE)小于叶绿素乙醇溶液总色差;叶绿素-酪蛋白分散体系在光照过程中zeta绝对值范围是30.6~38.2 mV。综上所述,酪蛋白酸钠包封叶绿素提高了叶绿素在水体系中的稳定性。该研究结果为拓宽叶绿素的应用提供了依据。

Research on the optimization of chlorophylls extraction and the stability of chlorophylls-casein in aqueous system
Abstract:Chlorophylls are the main pigments in photosynthetic plants,but their applications in food are limited due to their deficiencies of aqueous insolubility and lability to processing and storage conditions.These hindrances might be improved by sodium caseinate(NaCas),which structurally consists of both hydrophobic and hydrophilic groups.The single-factor and orthogonal test were used to optimize the extracting method of spinach chlorophylls and then spinach chlorophylls were fabricated in sodium caseinate using anti-solvent method.The stability of chlorophylls in casein dispersion was evaluated with chlorophylls remaining,color and electrostatic stability.The extracting amount of spinach chlorophylls reached the maximum of 1.89 mg/g when the extraction temperature was 50 ℃,ultrasonic power was 300 W,and extraction time was 30 min.The dispersion reduced light degradation of chlorophylls in aqueous dispersions due to the protective effect of NaCas.Chlorophylls in NaCas dispersion remained 62.96% after light treatment while chlorophylls in ethanol solution remained 56.07%.Additionally,zeta-potential of dispersions after light treatment was from-30.6 to-38.2 mV.This study showed that NaCas improved the stability of chlorophylls in aqueous system,which could potentially broaden the application fields of chlorophylls.
