VirB4缺失型特利波契巴尔通体入侵淋巴循环的研究

中文摘要:巴尔通体通过抑制宿主淋巴结内免疫反应,利用淋巴循环侵入血液导致持续性菌血症。 4型分泌系统是巴尔通体建立持续性感染的关键因素,但尚不清楚4型分泌系统在巴尔通体侵入淋巴循环中所起的作用。VirB4基因是4型分泌系统重要的组成部分,本实验以敲除VirB4基因的特利波契巴尔通体菌株与野生型特利波契巴尔通体菌株分别侵染大鼠淋巴循环系统来评价4型分泌系统对菌株侵入淋巴循环能力的影响。通过大鼠胸导管引流模型,用qPCR方法检测淋巴液中的菌载量。结果表明在不同的时间点,野生型与VirB4缺失型菌株均能够进入淋巴循环,但在感染6 h后,淋巴循环中的VirB4缺失菌数量大于野生型菌。提示4型分泌系统并不决定特利波契巴尔通体侵入淋巴循环的能力。

Investigation of invasion in lymphatic circulation by VirB 4 knock-out mutant of Bartonella tribocorum
Abstract:Bartonella is able to inhibit immuno-reaction in the infected lymph nodes.Consequently,Bartonella invade bloodstream from dermal inoculation by the transportation of lymphatic circulation.Type 4 secretion system is a key factor in the establishment of persistent infection of Bartonella,but it is not clear whether this apparatus determines the ability of Bartonella to invade the lymphatic circulation.VirB4 gene is an important part of type 4 secretion system.Here,wild type Bartonella tribocorum and VirB4 knock-out mutant strains were used to infect rats respectively.Whereafter,lymph samples were collected at different time points post injection through thoracic drainage surgery.The bacterial load in lymph fluid was detected by qPCR method.Results showed both wild-type and VirB4 deletion strains are able to enter the lymphatic circulation at different time points.However,the number of VirB4 deletion strain in the lymphatic circulation significantly increased more than wild type at 6 hours post infection.Results indicated that type 4 secretion system does not determine the ability of Bartonella tribocorum to invade lymph circulation.
